Progynon Depot (Estradiol Valerate)

  • Type: Hormone replacement therapy (HRT)

  • Main Ingredient: Estradiol valerate, an estrogen hormone

  • Form: Usually available as an intramuscular injection

How It Works

  • Estradiol valerate is a prodrug of estradiol — once injected, it is converted in the body to estradiol, the main natural female sex hormone.

  • Restores or supplements estrogen levels in the body.

  • Helps regulate menstrual cycles, relieve menopausal symptoms, and maintain healthy bones and reproductive tissues.

Common Uses

  • Hormone replacement therapy (HRT):

    • Relief of menopausal symptoms (hot flashes, night sweats, vaginal dryness)

    • Prevention of osteoporosis in postmenopausal women

  • Hormonal regulation:

    • Treatment of menstrual cycle disorders (amenorrhea, irregular cycles)

  • Part of hormone therapy:

    • Used in certain fertility treatments

    • May be used in gender-affirming hormone therapy (for transgender women, as prescribed)

Advantages

  • Provides steady estrogen levels because it is long-acting

  • Reduces symptoms related to estrogen deficiency

  • Supports bone density and cardiovascular health when used appropriately

  • Convenient dosing schedule — usually once every few weeks

Possible Side Effects

  • Breast tenderness or enlargement

  • Nausea or bloating

  • Headache or mood changes

  • Irregular vaginal bleeding (especially early in treatment)

  • Rare: blood clots, stroke, or increased risk of certain cancers (depends on long-term use and individual risk factors)

Precautions

  • Should be used under medical supervision, especially in women with a history of:

    • Blood clots

    • Breast or uterine cancer

    • Liver disease

    • Unexplained vaginal bleeding

  • Regular monitoring is recommended (pelvic exams, breast exams, and sometimes blood tests).

  • Usually combined with a progestogen in women who have not had a hysterectomy, to reduce the risk of endometrial hyperplasia (thickening of the uterine lining).
