Sargramostim Injection

Category: Hematopoietic growth factor / Colony-stimulating factor (GM-CSF)
Form: Injectable solution for subcutaneous (SC) or intravenous (IV) use

Uses:

  • Stimulates bone marrow to produce white blood cells, especially granulocytes and macrophages.

  • Used in patients with neutropenia after chemotherapy or bone marrow transplantation.

  • Can accelerate recovery of bone marrow function in certain cases of bone marrow failure or after autologous stem cell transplantation.

  • Sometimes used to mobilize peripheral blood progenitor cells for collection and transplantation.

Mechanism of Action:

  • Sargramostim is a recombinant human granulocyte-macrophage colony-stimulating factor (GM-CSF).

  • Binds to GM-CSF receptors on hematopoietic progenitor cells in the bone marrow.

  • Stimulates proliferation, differentiation, and activation of granulocytes, monocytes, and macrophages.

  • Enhances immune function and infection defense in immunocompromised patients.

Administration:

  • Administered subcutaneously or intravenously, depending on clinical protocol.

  • Dosage is based on body weight, clinical condition, and desired white blood cell response.

  • Frequency varies from daily to several times a week until hematologic recovery.

Side Effects:

  • Bone and muscle pain, due to increased marrow activity.

  • Fever, chills, fatigue, and headache.

  • Gastrointestinal: nausea, vomiting, diarrhea.

  • Cardiovascular: hypotension, edema (rare).

  • Hematologic: leukocytosis if overdosed.

Precautions:

  • Monitor complete blood counts during therapy to avoid excessive leukocytosis.

  • Use cautiously in patients with heart failure, pulmonary disease, or fluid overload.

  • Monitor for allergic reactions during injection.

  • Adjust dose in patients with renal or hepatic impairment if necessary.
